You will be required to know what a metabolic pathway is, and know that they are controlled by a series of enzyme based reactions.
If there is a mutation and one or more of these enzymes is not produced correctly (by protein synthesis) then it may mean the pathway cannot happen fully or even partially, this causes issues.
PKU is a disease caused as a result of the effect of a mutation on a metabolic pathway.
