World Journal of Microbiology and Biotechnology, 8, 564-566

Hydrolysis of raffinose and stachyose in

cowpea (Vigna unguiculata) flour, using

-galactosidase from Aspergillus niger

R.I. Somiari and E. Balogh*

Filamentous fungi, isolated from the enriched surface of garden soil, were screened for a-galactosidase production

after growing on wheat-bran/carbohydrate substrate. One isolate, Aspergillus niger, had the highest enzyme

activity (5.1 x 10 -2 units/mg protein) at pH 5.0 and 50~ Treatment of cowpea flour with the crude enzyme

reduced the raffinose and stachyose content by 95% and 82% respectively. This technique could therefore be

useful in controlling the flatulence activity of cowpeas.

Key words: Aspergillus niger, flatulence, 0r raffinose, stachyose.

R,I. Somiari and E. Balogh are with the Department of Food Science and

Technology, Obafemi Awolowo University, lie-Ire, Nigeria. *Corresponding

author.

ACKNOWLEDGMENTS

CytoPulse and University of Maryland, Baltimore, have had a collaboration

agreement under which CytoPulse’s equipment has been used by Robert

Malone, M.D., in scientific research and development, principally in the areas

of delivering DNA using pulsed electric fields. These studies were supported by

NIH-K02AI01370 (R.W.M.), NIH-R01RR12307 (R.W.M.), and developmental

funds provided by the University of Maryland Medical System (R.W.M.).
