Organ-Chip: Fabrication and Testing Work Flow
Hydrogel is made with A7r5 smooth muscle cell suspension and injected into FEP tube.
Immediately after hydrogel injected into FEP tube, 250 um wire is inserted within the tube.
After hydrogel cures, the wire is removed, leaving a "lumen" within the gel. This is then seeded with the epithelial (HUVEC) cells to emulate the environment of a blood vessel.
Artificial Blood Substitute (ABS) is then circulated through the lumen.
Cell response to ABS can be analyzed through metabolite collection and other measurables as seen on the left.

The organ-chip contains a tube based design to optimize lumen structural integrity and reduce the overall cost and use of materials.
Tube Design Materials:
3 mm diameter FEP tube
400 ul of Collagen Gel containing two cell lines
25 gauge needles for pumping
250 um wire for lumen fabrication
