Mayor Simplicio Manalo National High School is located at E. Quiogue St., Barangay Aguho, Pateros Metro Manila. It has an area of about Two Thousand Ninety Six (2,096) square meters lot donated by Late Col. Agripino Manalo in memory of their father who has been also the Mayor of Pateros, the respected late Agripino Manalo. This is also the very reason that the name of this school was being named after him.

This school was established through the effort of Congresswoman Ma. Laarni L. Cayetano and Sen. Alan Peter S. Cayetano. The bill for the creation of this school has been approved in 2008 by both the House of Representatives and the Senate as third public secondary school in the Municipality of Pateros. The approval of this measure further strengthens the united vision of providing quality education to the students of the Municipality of Pateros.

It was then that this school was established on December 2, 2010 in the middle of the School Year of 2009- 2010 in due respect to the request of the lot donor, Late Col. Agripino Manalo and his family to see the school operation while he still alive. And so that day and onwards is now a big history.

The establishment of this school brought about 335 students who were mostly from Barangay Aguho and other adjacent barangays. The teaching staff for the first phase of implementation was selected from teachers at Pateros National High School. The school temporarily utilized the eight vacant rooms of Aguho Elementary School while the construction of the school building is on-going. The mission of the lot donor is to establish this school intended solely for High School students.

On June 23, 2014, all students and teachers were transferred at the new school building at 3 storey building with 20 classrooms that is now operational. This is now the school owned building that caters grades 7 to 10 students

On June 13, 2016, the school also started Grade 11 for the Senior High School program while waiting for their own seperate building. There are 26 students in Technical Vocational & Livelihood Course who were enrolled in the first semester of School Year 2016 – 2017. Some teachers from Junior High School also handle classes in Senior High School.
