Over the past 30 years, transition metal compounds have shown great medicinal promise because of their diverse structures, chemical stability, and electronic tunability. One such complex is cis-platin (Figure 1).
Cis-platin is used extensively to combat many forms of cancer and in particular, it has been used to fight testicular cancer with a 95% survival rate. Unfortunately, this therapeutic, like many others, has severe side effects including hair loss, nausea, and the loss of taste. These problems arise because the drug attacks both healthy and cancerous cells.
Recent studies have shown that cancer cells have an over expression of folate receptors. As a result, folic acid (Figure 2) is preferentially taken up by cancerous cells, and hence, this moiety may be used to selectively target tumors while leaving the health cells alone.
For the past several years, the Krogstad research group has extensively examined the water-soluble phosphine 1,3,5-triaza-7-phosphaadamantane (PTA, Figure 3), its derivatives, and their metal complexes.
This summer, we plan to prepare new folic acid derivatives of PTA and to coordinate the water-soluble phosphines to Ru and Pt precursors. It is hoped that the resulting Pt and Ru compounds (Figure 4) will selectively target cancerous cells, and therefore have limited side effects for the patient.
This will involve utilizing synthetic methodologies from both organic and inorganic chemistry.
