Autoclave Rotation
Duties:
Gather all glassware from drying areas and prepare for autoclaving (i.e., wrap bottles/beakers caps with tin foil and stick a piece of autoclave tape).
If possible, try to reserve a time slot in the autoclave paper sheet (just write a “R” in the comments section).
When the autoclaving process has finished, please put the material back into the corresponding cabinets.
It is mandatory to autoclave during office hours (9:00 AM-6:00 PM). In case something goes wrong there will be someone to assist.
Important notes!!!
Wash and dry glassware are up to the end user. Please rinse thoroughly if you are washing with detergent or your solution contained detergent (If you see small bubbles after rinsing, it means there are still traces of detergent).
If you don’t know if a labware is autoclavable, please ask to somebody.
C. elegans Buffer Prep Rotation
Duties:
Responsible for checking and preparing the buffer stocks designated in your rotation schedule.
Responsible for removing the full waste box in the worm room by following the rotation schedule.

Ask someone if you are not sure how to prepare them!
Buffer and Solution List:
1 M CaCl2
Make a new aliquot of 50 ml solution if only 2 tubes are left.
*1 L = 20 tubes of 50 ml aliquots
1 M MgSO4
Make a new aliquot of 50 ml solution if only 2 tubes are left.
*1 L = 20 tubes of 50 ml aliquots
2X Freezing Solution
Make a new aliquot of 100 ml solution if only 2 bottles are left.
*1 L = 10 bottle of 100 ml aliquots
B. Broth
Make a new aliquot of 100 ml solution if only 2 bottles are left.
*1 L = 10 bottle of 100 ml aliquots
Cholesterol stock
Make 50 ml of new stock solution (5 mg/ml) if only 1 tube is left.
*Make 2 tubes
KPO4 stock
Make a new aliquot of 200 ml solution if only 2 bottles are left.
*1 L = 5 bottle of 200 ml buffer
M9 Buffer
Make a new aliquot of 125 ml solution if only 2 bottles are left.
*1 L = 8 bottle of 125 ml buffer
NGM agar plates
*Refer to plate pouring schedule
General Buffer Prep Rotation
Duties:
Responsible for checking and preparing all the buffer stocks which are listed below on the week of your rotation schedule.

I arrange this into the monthly rotation since most of the buffer only needs to be prepared at least once a month.
Buffer and Solution List:
Ethanol 70% for disinfectant
Make 5 L of ethanol 70% if only ¼ of the volume is left.
LB Broth
Make a new aliquot of 250 ml solution if only 2 bottles are left.
*1.5 L = 6 bottle of 250 ml buffer
LB-Carbenicillin Agar plate (50 μg/ml Carbenicillin)
Make a new batch of antibiotics plates if only 5 plates are left.
*2 L = 80 plates. Don’t make more than this, because the antibiotic plate has to be used as soon as possible (not more than 2 months if possible).
100 mg/ml Carbenicillin stock
Make a new aliquot of 1 ml stock if only 2 tubes are left.
*20 ml = 20 tubes aliquots
10X Tris-Glycine buffer (Running Buffer for western blot)
Make 5 L of 10X Tris-Glycine buffer if only ± 1 L of the volume is left.
*Do not mix new buffer with old buffer. Put the old buffer into a glass bottle and label it.
