The show's theme song features the voices of all six leading characters, and is played in every episode's title sequence. Shortened instrumental versions of the theme song play on the menus of the show's DVDs, and modified versions of the theme song play in some of the brand's toy commercials. The song is also included in the My Little Pony Live Show.

Except Italian season one[note 1] and the Japanese broadcasts on TV Tokyo which both have their own themes, most of the non-English versions, including Boomerang and Cartoonito Italy's broadcasts of subsequent seasons, as well as Dlife and Disney Channel Japan's reairings of the first two seasons, use translations of this song or keep the song in English.

A remastered version of the theme song debuted in Lesson Zero.[2] It features unaltered lyrics, but Rebecca Shoichet's vocals for Twilight Sparkle are re-recorded, the instruments are higher in the mix in relation to the vocals, it features a rising "whoop" sound as Rainbow Dash spins through the clouds, and it is accompanied by a slightly different animated sequence. "Creme Brulee" is replaced by Big McIntosh, Granny Smith walks in the opposite direction, there is an addition of a train station and a steam-engine Friendship Express train running through Ponyville, with Derpy peeking from one of the cars' windows and the Cutie Mark Crusaders sitting in the last car, and Princess Celestia's magic now has its proper yellow glow. The melody of the first theme can still be heard in the end credits for most episodes.

During "it's an easy feat, and magic makes it all complete", Twilight briefly flies in front of the castle before sitting with her friends, the Young Six, and several background students and posing for a class photo taken by Photo Finish. After the camera flash, Spike uses his fire breath to send the photo to Canterlot, but the castle throne room now matches its appearance from My Little Pony The Movie, with two thrones for Celestia and Luna. Celestia and Luna appear with Princess Cadance, Shining Armor, and Flurry Heart when the photo arrives. They look at the photo together, and the theme song ends on the show's logo.

Fluttershy does this (and adds a few more bars from the 1980s theme) in The Ticket Master when she and her animal friends are cleaning the Golden Oak Library for Twilight, and again in A Bird in the Hoof, harmonizing with her hummingbird friend Hummingway. Spike hums the tune in Griffon the Brush Off when he carries the scrolls out of the town hall before Pinkie Pie and Rainbow Dash scare him. The bat in May the Best Pet Win! plays the tune on a glass harmonica. S04E08 Unnamed Earth Stallion #1 and "Tornado Bolt" play the tune on a giant piano at a Manehattan toy store in The Gift of the Maud Pie. Russell Ferguson sings along with the song at the Golden Oak Library in a Hub Network commercial.[3]

The theme song has a roughly two-minute extended set of lyrics. A small portion of the extended version of the song was originally introduced as one of three songs included with the My Little Pony Twilight Sparkle Animated Storyteller, an animatronic, plush toy in the form of Twilight Sparkle.[6] An instrumental version of an earlier portion is played in My Little Pony Friendship is Magic: Discover the Difference.

The opening rhythm in the song echoes the classical My Little Pony theme. The original 10-note theme that is heard in all "My Little Pony" themes was written by Anne Bryant, Clifford "Ford" Klinder, and Barry Harman.[8] Lyrics sung by the other main characters are not included in the extended version.

However, I did more searching around on the origins of the My Little Pony Theme Song, and I found an even older instrumental containing a guitar riff that sounds like it evolved into the first few notes of the theme song. The theme song of tag_hash_112______________ generally has the same two tunes from G1 to G3, though there are many variations. Although originally dating back to the 1980s with MLP G1, the theme song's first appearance in G3 is in A Charming Birthday where it is used in the end credits. It is then subsequently used in the end credits of the other movies but lyrics are added for My Little Pony Live: The World's Biggest Tea Party live show. Later on, a new theme is added to commemorate the Core 7 era.

This theme song is a song modified from past versions of the theme song from previous generations. It first appears as a rock and roll tune without lyrics during the dance montage in A Charming Birthday; a more soothing version is used in the end credits, as well as at the end of each movie thereafter.

For the Core 7 era, the song is modified to coincide with the changes to the toyline. This version of the theme song weaves all 7 ponies together and uses the lyrics to showcase their activities. The extended versions includes more general MLP-based lyrics and some that are modified from the original Core 7 version of the theme song.

As you can see in the file I attached, Discord's in the theme song. I'm pretty sure this is just in Season 4, and might only be in Trade Ya (I haven't checked yet, I posted this as soon as I saw it when watching the new episode, Trade Ya).

Only in season 1, the songs of the dub were performed by voice talents in Malaysia, instead of the dub's home country of Indonesia. Despite this, this version of the theme would be kept for 6 more seasons.

This version has been used on TV only from season 2 onwards, as season 1 had its own theme song. Releases of the show on external media like DVD, Netflix or YouTube use this theme song on season 1 episodes too.

The Japanese theme songs had their own original arrangements in the first two openings, which can be seen here. Since season 3, and in Disney-owned channels' reruns of the first two seasons, the opening has been a direct Japanese dub of the original theme song.

Hasbro allegedly told the staff of the Korean dub of the series that they were not allowed to dub the songs, for unknown reason. The staff negociated with Hasbro that if any dialogue comes before the song, then it is okay to dub it. Therefore, the opening theme is one of a few songs that were not dubbed since the dub began.
