If you just want to cut together an entire song from a master clip that has several takes on it, you'd just start by marking the first part of the master clip you want to put in the sequence and edit it in. Then mark the next part and edit it in right after the first part. Keep going until the song is done.

If you set up masters for individual question types, they won't be applied when adding questions of those types - it'll always default to the main question master and then as you mentioned you can change the layout.
