I seek to identify the molecular mechanisms instructing astrocyte generation in the developing mouse and human cerebral cortex. Among the major developmental signalling pathways, Wnt/β-catenin has been shown to play highly diverse roles in the formation of the brain, depending on developmental time and spatial position. I seek to investigate whether Wnt/β-catenin signalling could play a role in regulating astrocyte differentiation in the neocortex.
Activation of the WNT/β-Catenin pathway leads to the translocation of β-Catenin to the nucleus, where it can subsequently bind to LEF1/TCF transcription factors (LEF/TCFs) and activate target genes. The LEF1/TCFs transcription factor family is composed of four proteins named: LEF1, TCF7, TCF7L1, and TCF7L2. My results as well as recent RNASeq data suggest that β-catenin and its effectors are expressed in astrocytes and in their precursors.
The aims of my study are to investigate at which stages of astrogenesis WNT/β-catenin effectors are expressed and how this pathway influences astrocyte function and development in the neocortex.
