Lore
Vampires were originally a group of nobles who drunk the blood of their enemies to get stronger. The blood magic turned these nobles into demonic batlike creatures called vampires.
These vampires conquered the world and ruled all the kingdoms until the Hunters rose up to stop them. Hunters are a threat because they can use garlic injections to prevent vampires from feeding.
As a vampire, you can stay out of geopolitics or get involved in restoring vampire glory by conquering villages (click the beacon in center of town to start a vampire invasion)

The vampires conquered the world and ruled all the kingdoms. These vampires feed on the underclass until a group known as the Hunters discovered that vampires have many weaknesses.
Hunters injected themselves with garlic powder so they'd be able to resist the deadly effects of vampire bites.

Lore
Werewolves recently got involved in the war between vampires and hunters. They are a third faction of humans bitten by wolf beasts. The vampires are a threat because they consume animals the werewolves rely on. The hunters are a threat because they don't like their livestock being eatten. The werewolves must therefore defeat these factions to carve out their own destiny

Normal player during the day (that means you lose werewolves perks but unlike vampires you aren't weakened by daylight)
You're weak to wolvesbane plants, those can be put in a diffuser making a chunk poisonous
You're minorly weak to water, you get weakness but it doesn't actually hurt you

The vacant are those who aren't willing to pick a side in the conflict, those who avoided being bitten, or those who are too proud to inject themselves with random stuff.
How to Join
Don't use any injections or vampire fangs
If you ever get turned into a vampire/werewolf, inject yourself with garlic
Get garlic from villager trades/structures around the world

You MIGHT be able to hold your own in the day against a weak werewolf or somewhat strong vampire
Hunters are basically a buffed version of you so regardless of day or night you'd have trouble with them
You can betray your Vampiric War Faction
Vampires, werewolves, hunters, and humans aren't strict factions, rather they're races which generally align with their own kind.
Betrayals during Village Raids
You could help another faction take over a village but...
I wouldn't recommend it since you'll lose out on villager trades and there will be hostile guards at the village conquered
Multi Species Nations/Towns
Nations/Towns can have multiple species or they can make prejudice laws barring species from membership/rights
Check in with a nation before you join them
FYI Nation Leaders ; you SHOULD tell people if you're prejudice against certain species
