A. 核酸適體aptamer的篩選方法研究
進行aptamer篩選方式改良,導入自動化、修飾鹼基等方式,增加篩選效率並提升篩選結果之親和力,以創造更多應用價值。另外使用核酸的conformational switch原理,以進行小分子aptamer的篩選開發。
B. 核酸檢驗試劑開發
鑽研各項核酸放大技術,配合不同探針設計,在不同平台上開發針對癌症突變、罕見疾病及感染症的核酸檢驗。包括:
(1) 基於PCR核酸放大的檢測方法:新型PNA, TFO探針開發與應用
(2) 基於恆溫核酸放大的檢測方法:應用現今較成熟之恆溫放大技術,開發簡便實用的核酸快篩。
(3) 檢驗醫學與工程的結合:配合高靈敏度的生物感測器開發新型核酸檢驗、結合微流道技術開發Lab on a chip (與工學院的合作計畫)
C. DNA奈米科技的應用
探索核酸結構互動與行為,以核酸作為積木拼裝,組成可編程分子電路,並將其應用在科學研究與醫學檢驗上。例如 PER、HCR
