Novel Cathepsins

9 out of the 11 human cathepsins have been expressed as active proteases in various expression systems.  So far only cathepsins O and W have evaded recombinant expression and enzymatic  characterization.  Our aim is to express both enzymes either in a yeast or mammalian expression system and to characterize their substrate and inhibitor specificity. The flowchart below depicts our approach for the expression and characterization of cysteine proteases.
