I would like to discuss our recent study that was published in the journal of Current Pharmaceutical Biotechnology. This study's title is “a survey and a molecular dynamics MD study on the central hydrophobic region of prion protein”.
•The central hydrophobic region of PrP includes a palindrome and a 3 repeated GxxxG motif.
•Firstly, we did a detailed survey on this region for the last 28 years and especially for the last 5 years.
•Secondly, in the PDB Bank, we already have the NMR structure of this region, the PDB ID is 2LBG.We did MD study for the wild-type and its 17 mutants.
• From the MD secondary structures and snapshots, we know all these 18 models start to unfold from the second nanosecond.
•We can observe the following 5 points: (1) The NMR structure short α-helix can be broken into 2 parts, usually just from the mutation point. The first half is very unstable, but the second half is much more stable than the first half, due to 3 hydrogen bonds always there.(2) AAAA, even the palindrome AGAAAAGA, is the most hydrophobic core. (3) HIS111 is a residue that plays important roles.(4) The 3 repeated GxxxG motif can inhibit prion infection.(5) The AGAAAAGA palindrome is very interesting to formulate a special ‘cage’ due to its hydrophobic property.
