G-M406

This is the home page for men who have the M406 mutation and those predicted to have
it.  This includes men who have not tested for all the known M406 subgroups.  For those 
men who are only predicted M406 or were not tested for M406 subgroups, additional 
testing is always most helpful.

Also included here are men from five subgroups of M406 whose ancestors developed a
a specific marker value oddity that has been passed down.  Because this is not 100%
reliable, these subgroups will be moved to their own pages when whole sequencing
describes a permanent SNP mutation they share.  

M406 has 4 subgroups defined by permanent SNP mutations, the L14, L645, PF3296
& M3302 subgroups.    M406 also has two major marker subgroups, DYS594=8
and DYS436=14.   As of this writing, neither has been tested for the official subgroups and may or
may not fall within a SNP-defined subgroup of M406.

The above diagram shows M406's position in the G tree.  It is the youngest of a
series of mutations, each one covering fewer persons as the tree branched.
All M406 men have all the mutations shown above it in the tree.

M406 has one sister subgroup, CTS5762.   Men can belong to one of them, but not to the
other because each has some different ancestors.

M406 men of all types often have the value of 21 at marker DYS390.  This is not 
a perfect identifier of M406 men because 21 occurs in small numbers in other G men
but can be helpful.  M406 seems to have been identified first in 2004 in a study by
Cinnioglu et al.

Age of M406

In the 2012 study by Rootsi et al., they indicated "The expansion time of G-M406 in 
Anatolia is 12,800 years ago, which corresponds to climatic improvement at the beginning 
of the Holocene and the commencement of sedentary hunter-forager settlements
at locations, such as Gobekli Tepi in Southeast Anatolia,  thought to be critical for the
domestication of crops (wheat and barley) that propelled the development of the 
Neolithic."  They also indicated  that M406 in Italy has a Td [coalescent time] of 
8100 years ago, "consistent with the model of maritime Neolithic colonization of the Italian
peninsula from coastal Anatolia and/or the Levant. Finally, to the east, G2a3a-M406 has 
an expansion time of 8800 years ago in Iran, a time horizon that corresponds to the first 
Neolithic settlements of the Zagros Mountains of Iran."
But there are problems with these dates.  They seem to presume that M406 moved to
the areas mentioned at the time mentioned.  It is also possible the men lived in a small group
elsewhere, and then migrated there in a more recent period.  In addition, the method used
to calculate these dates relied on the number of marker mutations.  It seems that counting
SNP mutations may be more reliable, and whole genome sequencing should soon provide
its own set of time calculations.  The coalescence time they list for the Caucasus is 9,200 yrs
and 8,800 yrs for Iran, 8,000 yrs for Italy and only 1,900 yrs for the Druze of the Levant.

Francalacci et al. in their study of sequenced samples in Sardinia provided information that
suggests the common male ancestor of the M406 men living there lived about 8,600 yrs
ago using the most widely used Y-DNA mutation rate.  This does not mean they were 
living there that long.

As to the DYS594=8 subgroup, comparisons of 67 markers suggest this subgroup may
share a common ancestor who lived 2000-3000 yrs ago.    The DYS436=14 subgroup seems
to have a common male ancestor a bit older.  

Geographical Distribution of M406

M406 men from the major subgroups are found in Europe, the Middle East and countries
south of the Caucasus Mountains.   Possibly of significance, likely M406 samples of any
type have not surfaced in central and southern Asia and western China though other G
subgroups are found in those locations.  In addition, M406 of all types seems even less
common in the far north European countries than other G subgroups.

In the 2012 study by Rootsi et al., they indicated "G-M406 has a peak frequency in
Cappadocia, Mediterranean Anatolia and Central Anatolia (6–7%). ....M406 has a modest 
presence in Thessaly and the Peloponnese (4%), areas of the initial Greek Neolithic 
settlements." (citing the 2008 study by King et al.)  They also indicated M406 occurs in 
Italy (3%).  These figures are based on the population of men of all haplogroups.
One interesting finding is that among the Druze of the Levant they found 25 M406+
samples out of 30 G samples, which is a huge percentage.  Earlier studies found 
Druze were less than 15% G overall.  http://en.wikipedia.org/wiki/Druze  These Druze have
an atypical 22 at marker DYS390 and were found negative for L14.

The DYS594=8 subgroup is very small so far.  There is one man from the Ukraine and
a Greek surname.  The DYS436=14 subgroup is also small and lists Czech, Saudi and
Spanish origins.  

Using DYS390=21 as an imperfect identifier of M406, the percentage of M406 in G men can 
be estimated in various locales.  These are M406 numbers per likely G men at various sites,
mostly from samples in the YHRD public database:

One conclusion can be made is that the percentage of M406 drops in northern Europe.

Balanovsky et al. in 2011 sampled 1525 in the north Caucasus area and found only one
man M406+.  This is an area with high concentrations of other G subgroups.

Francalacci et al. in 2013 showed that M406 men (seemingly negative for L14 and 
L645) represent about 10% of the G men in Sardinia.

Origins of M406

Everything available presently seems to indicate that the M406 mutation occurred
before the period in which records were kept.  This being the case, the early M406 men
could have been living most anywhere.

About 2010 we polled the samples in the G project and found the following M406 
concentrations among customer samples in Italy 20% of 156 G samples......
Spain 15% of 56 G samples.....Netherlands 15% of 20 G samples......
Switzerland 8% of 51 G samples.....Iran 6% of 34 G samples....Poland 4% 
of 75 G samples......France 4% of 46 G samples......Ireland 3% of 29 
G samples.......India 3% of 18 G samples.

Maps Related to M406

On the Project Roster, subgroups have their own maps.   
Look at top left of map and choose the M406 subgroup.  Just below this are the maps for the 
subgroups of M406.

This map (at bottom) from a 2012 study by Rootsi et al.
provides a M406-only map, but it is very tiny.  Magnification to 200% is suggested.
This suggests an unusual M406 concentration is found only in inland Turkey.  But
data they did not use suggests a higher concentration in Greece and northern Iraq.
And a different percentage scale would have made the next most concentrated M406
areas along the northern Mediterranean more visible on the map.
