Ligation
Protocol
Clean up restriction enzyme-digested DNAs (i.e., the vector and the insert)
Run gel to determine the "molecular ratio" of DNAs
Ligate the vector and the insert at a ratio of *1:15-20 with T4 DNA ligase in the ligation buffer (20 ul).
Incubate at room temperature for 4 hr.
Subject to performing Transformation.
