Signalling molecules in Hippocampal slice Neurodegeneration associated with CNS diseases potentially involves the dysregulation of vital signalling pathways within the cells of the brain. Utilising models of CNS disorders, we can mimic neurodegeneration associated with stroke and Multiple Sclerosis and investigate how the signalling pathways are altered under these conditions. Using powerful microscopy methods, we can identify neurones (stained green) and the levels of a signalling molecule called ERK (stained blue) and how much of this is active (phospho-ERK, stained red). Understanding how signalling molecules are altered in our models of CNS disorders will lead to a greater understanding of their contribution to CNS diseases and how we may target these for therapeutic purposes. Technical details : Image shows the CA3 region of an organotypic hippocampal slice labelled with antibodies against MAP2 (a somatodendritic neuronal marker; green), ERK (an intracellular signalling molecule; blue) and phospho-ERK (the active form of ERK; red), against which appropriate Alexa Fluor-conjugated secondary antibodies were directed. Neuronal processes containing activated ERK appear white. Image was captured using the Leica SP5 Confocal system with a 40x oil immersion objective using excitation wavelengths of 405 nm, 488 nm and 633 nm. I discovered this two-way radio in the basement beneath the "Reunification Palace" in Ho Chi Minh City, Vietnam. I love the big dial in the middle that measures "Excitation". Wonder what that was for? Prior to the reunification of Vietnam, the Palace was know as the Independence Palace and was the residence of the president of South Vietnam. Construction of the building started in 1962 after the previous residence on the site was bombed by South Vietnamese airforce pilots in an attempted Coup d'etat. Needless to say, the new building was built with large and secure bunker underneath, where I found this two-way radio.
